Dissolved Oxygen (DO): Definition, Importance, Factors & Measurement Methods

Dissolved Oxygen (DO): Importance, Measurement Methods & Winkler Method Explained.

What is Dissolved Oxygen (DO)?
Dissolved Oxygen (DO) is the amount of oxygen dissolved in water. It is one of the most important indicators of water quality because aquatic organisms such as fish, insects, and other aquatic life depend on dissolved oxygen for survival. If the DO level becomes too low, water bodies become unsuitable for aquatic organisms and overall ecosystem health declines.
DO is widely used to assess the quality of natural water and wastewater.
Why is Dissolved Oxygen Important?
Dissolved Oxygen is essential because it:
Supports the survival of aquatic organisms.
Indicates the health of rivers, lakes, ponds, and streams.
Helps monitor pollution levels in water.
Is a key parameter in environmental monitoring and wastewater treatment.
Reflects the biological condition of a water body.
Factors Affecting Dissolved Oxygen
Several natural and environmental factors influence the dissolved oxygen concentration in water, including:
Geographical terrain
Water inflow and outflow conditions
Physical properties of water
Chemical composition
Biological activity
Temperature
Organic pollution
Methods for Measuring Dissolved Oxygen
The commonly used methods for estimating Dissolved Oxygen are:
Membrane Electrode Method
Electrochemical Method
Winkler Iodometric Method
Azide Modification of Winkler Method
Among these, the Azide Modification of the Winkler Method is the most widely used because it provides reliable and accurate results while reducing interference from nitrite ions.
Azide Modification of the Winkler Method
This method is based on a chemical reaction that converts dissolved oxygen into an equivalent amount of iodine, which is then measured by titration.
Principle
Manganous sulphate reacts with an alkali (NaOH or KOH) to form a white precipitate of manganous hydroxide.
In the presence of dissolved oxygen, the precipitate is oxidized into a brown-colored compound.
After adding acid, iodide ions reduce the oxidized manganese compound and release iodine.
The released iodine is directly proportional to the dissolved oxygen present in the sample.
The iodine is titrated with sodium thiosulphate using starch as an indicator.
The azide modification eliminates interference caused by nitrite ions, improving the accuracy of DO determination.
Applications of Dissolved Oxygen Measurement
DO measurement is widely used in:
Environmental monitoring
Drinking water quality assessment
Wastewater treatment plants
Fisheries and aquaculture
River and lake pollution studies
Scientific research
Conclusion
Dissolved Oxygen is one of the most critical parameters for determining water quality. It directly affects aquatic life and serves as an indicator of environmental health. Among the various measurement techniques, the Azide Modification of the Winkler Method remains one of the most accurate and widely accepted methods for estimating dissolved oxygen in water samples.
